For the visit of Manchester United, Ajax will have to do without several members of the first-team squad as injuries continue to take their toll.
In defence, Andre Ooijer (calf) won't be involved, neither will Gregory Van der Weil (groin), with youngster Dico Koppers likely to start at left-back.
In midfield the hosts have mixed news as Theo Janssen (groin) is ready to return and could even start, but Eyong Enoh (hamstring) and Derk Boerrigter (back) look very doubtful, while in attack Kolbeinn Sigthorsson (back) is struggling to be fit.
United's injury problems have been made worse with the news that Ryan Giggs, Dimitar Berbatov and Patrice Evra all miss out on the trip to Holland. Tom Cleverley (ankle), however, could start after taking a place on the bench against Liverpool, while Nani (ankle) and Phil Jones (foot) may also be able to feature.
Chris Smalling (slight knock) remains touch-and-go for the trip to Amsterdam, while Anders Lindegaard (ankle), Anderson (unspecified knock) and Michael Owen (thigh) remain a couple of weeks away from action.
Two clubs with seven European Cup successes between them go head-to-head in the Europa League, with Ajax set to play host to Manchester United.
Continental giants from Holland and England will lock horns in the last 32 of a competition both would rather have avoided.
Shock eliminations at the group stage of the UEFA Champions League have forced Ajax and United into Thursday night football, with both having laid out much grander plans before the season got underway.
They are, however, determined to make the best of the situation, and both clubs have voiced their desire to go all the way in the Europa League.
Ajax are one of only three sides to have tasted European Cup/Champions League, UEFA Cup/Europa League and Cup Winners' Cup success, while United are looking to join that exclusive club this season.
Ajax's record against English opposition is not the best, with seven outings having come and gone since they last tasted victory, and they will be aware that United will pose another considerable test in midweek.
The Red Devils have won six of their previous 10 outings against Eredivisie sides, but have only secured one victory in five on Dutch soil.
The reigning Premier League champions find themselves absent from the knockout stage of the Champions League for the first time in six years and are set to take in UEFA Cup/Europa League action for the first time since 1995/96.
They have only been involved in four UEFA Cup fixtures under Sir Alex Ferguson, and all of those games finished in draws.
Ajax and United have only met on one previous occasion in European competition, with Tommy Docherty's Red Devils securing a 2-1 aggregate success in the first round of the 1976/77 UEFA Cup.
Manchester United will rest Patrice Evra, Ryan Giggs and Dimitar Berbatov for their Europa League last-32 first-leg tie at Ajax.
Manager Sir Alex Ferguson said Evra would be rested in able to recover from events during United's victory over Liverpool at the weekend.
Winger Nani and defender Phil Jones have been named in the 20-man squad having recovered from injury.
Midfielders Ashley Young and Tom Cleverley have also travelled.
Youngsters Paul Pogba and Zeki Fryers are also in the squad for the match in Amsterdam.
Despite the pedigree of the opposition, Thursday's game is the first time United have competed in Europe outside the Champions League since 1995.
On that occasion they were beaten on away goals by Russian side Rotor Volgograd in a Uefa Cup first round tie.
United are joined in this season's Europa League by rivals Manchester City, whom they could face in the quarter-finals if they beat Ajax and City get past holders Porto.
United striker Javier Hernandez admits the Red Devils would have preferred to be participating in the Champions League this week, but says the stature of their opponents means the game retains some prestige.
"Both teams have come out of the Champions League and they both have a great history," said Hernandez. "We have won a lot of European Cups between us, so it will be a very interesting game.
"We need to get used to the fact we are not in the Champions League this season."
Despite not being in the elite competition of European football for the remainder of the season, manager Sir Alex Ferguson is hoping his side progress in the Europa League in order to have a chance of winning it.
"Winning any European trophy is significant. Irrespective of those who have won the three trophies before it is important for us, with our own history, to win tournaments all the time," he said.
"We threw it away in our home game against Basel and were unlucky against Benfica. That caught us short and we suffered for it.
"Now players are coming back I am sure we can combine the Premier League and Europa League so I would certainly try to play my strongest team in each round now."
Manchester United squad: De Gea, Amos; Fabio, Rafael, Evans, Jones, Smalling, Fryers, Ferdinand; Valencia, Nani, Carrick, Young, Park, Scholes, Cleverley, Pogba; Rooney, Hernandez, Welbeck.
